چکیده

Psoriasis (PsO) is a T cell-mediated autoimmune disease without cure. The major risk-associated PsO susceptibility gene encodes for the human leukocyte antigen C*06:02 (HLA-Cw6). Half of patients carry this allele encoding HLA-I molecule which presents short peptides (together known as immunopeptidome) to CD8 cells, thereby initiating immune responses. It unclear presented by HLA-Cw6 are prevalent trigger PsO. We generated immunopeptidomics data from skin-derived HLA-Cw6+ cell lines (>24,000 peptides) and newly HLA-Cw6Tg mouse model in conjunction with imiquimod-induced (>4,000 thus identifying set high relevance finding universal molecular triggers Strikingly, 58% characteristic 9-mer exclusive skin, were either identical or very similar (1 amino acid change) that we detected melanocyte/keratinocytes. To interrogate – interactions, performed single receptor sequencing clonal analysis skin cells lesional biopsies HLA-Cw6+/HLA-Cw6- patients. currently screening these receptors against our candidate establish dominant peptide broadly recognized autoreactive cells. conclude applying discovery new preclinical HLA-Cw6Tgmouse valid means study PsO-relevant immunopeptidome. Defining HLA-Cw6-restricted epitopes PsO, will facilitate therapeutic approaches develop tolerisation strategies potential achieve long-lasting remission.

Melanocyte antigen triggers autoimmunity in human psoriasis

Psoriasis vulgaris is a common T cell-mediated inflammatory skin disease with a suspected autoimmune pathogenesis. The human leukocyte antigen (HLA) class I allele, HLA-C*06:02, is the main psoriasis risk gene. Epidermal CD8(+) T cells are essential for psoriasis development. Functional implications of HLA-C*06:02 and mechanisms of lesional T cell activation in psoriasis, however, remained elus...

Diet-induced hepatocellular carcinoma in genetically predisposed mice.

Hepatocellular carcinoma (HCC) is one of the leading causes of cancer death worldwide, with approximately 70% of cases resulting from hepatitis B and C viral infections, aflatoxin exposure, chronic alcohol use or genetic liver diseases. The remaining approximately 30% of cases are associated with obesity, type 2 diabetes and related metabolic diseases, although a direct link between these patho...

Are elite endurance athletes genetically predisposed to lower disease risk?

We compared a polygenic profile that combined 33 disease risk-related mutations and polymorphisms among nonathletic healthy control subjects and elite endurance athletes. The study sample comprised 100 healthy Spanish male nonathletic (sedentary) control subjects and 100 male elite endurance athletes. We analyzed 33 disease risk-related mutations and polymorphisms. We computed a health-related ...

Is Post-ERCP Pancreatitis a Genetically Predisposed Complication?

Background/Objectives. Pancreatitis remains the most common complication of ERCP. History of post-ERCP pancreatitis is an independent risk factor for a new episode, suggesting a genetic background. The N34S mutation in serine protease inhibitor Kazal type 1 (SPINK 1) gene may downregulate the threshold for the development of pancreatitis. The aim of the present study is to evaluate the presence...

Staphylococcus Aureus Carriage in Patients with Psoriasis

Background: The aggravating role of Staphylococcus aureus is well known in atopic dermatitis but has not yet been proven in psoriasis. The role of Staphylococcus aureus superantigens is emphasized in the initiation, maintenance and complications of psoriasis. We investigated the frequency of nasal, axillary, and perineal carriage of Staphylococcus aureus (SA) in patients with psoriasis and its ...
